چکیده

The perivitelline space is the gap between the plasma membrane and the zona pellucida of the oocyte. Although not present in the oocytes in the germinal vesicle stage, the perivitelline space is known to start forming when meiotic maturation resumes and to enlarge in matured oocytes at the metaphase II stage (Kaufman et al., 1989; Okada et al., 1993). Recently, it has been suggested that there is a relationship between the size of perivitelline space in porcine oocytes and the incidence of polyspermy in those following insemination (Funahashi et al., 1994; Wang et al., 1998), but it is not clear whether the relationship is present in oocytes of another animals. It is generally thought that glycoproteins synthesized and secreted by cumulus cel ls are involved in the enlargement of the perivitelline space (Talbot and Dandekar, 2003). Glycoproteins secreted from cumulus cells accumulate between cumulus cells and in the perivitelline space (Talbot, 1984). Hyaluronan, a major component of the glycoprotein synthesizd and secreted by cumulus cells, in particular has a water absorbing property which results in enlargement of the matrix of cumulus and the perivitelline space (Talbot and Dandekar, 2003). In the cumulus-oocyte complex (COC) with cumulus cells whose glycoprotein synthesis was inhibited, therefore, it is supposed that the perivitelline space in oocytes fails to enlarge and the incidence of polyspermy after insemination increases. In the present study, the size of the perivitelline space was measured in mouse oocytes treated with tunicamycin, an inhibitor of glycoprotein synthesis, and also the incidence of polyspermy in those following insemination was observed, in order to examine whether the relationship between the incidence of polyspermy and the size of perivitelline space exists in mouse oocytes.

Comparison of the Size of Perivitelline Space in Mouse Oocytes Cultured with and without Cumulus Cells and Possible Hyaluronan Production in Oocytes

Perivitelline space is the gap between the plasma membrane and zona pellucida of oocytes. Recently, it has been reported that the incidence of polyspermy in porcine and mouse oocytes with larger perivitelline space is significantly lower than those with small perivitelline space (Funahashi et al., 1994; Wang et al., 1998; Kitagawa and Niimura, 2006; Ueno and Niimura, 2008). Therefore, it has be...

Perivitelline space: does it play a role in blocking polyspermy in mammals?

The perivitelline space of mammalian oocytes changes in size and composition during preimplantation development. Often overlooked in the past, this space contains a hyaluronan-rich extracellular matrix prior to fertilization and a cortical granule envelope following release of the cortical granules at fertilization. The hyaluronan-containing matrix of unfertilized oocytes is well developed in s...

Sericin Accelerates the Production of Hyaluronan and Decreases the Incidence of Polyspermy Fertilization in Bovine Oocytes During In Vitro Maturation

Fetal bovine serum (FBS) has been widely used as a supplement in the maturation medium of bovine oocytes in vitro. However, serum contains many undefined factors and is potentially infectious to humans and animals. As a serum replacement, we evaluated the feasibility of using the silk protein, sericin, derived from the cocoons of silkworm. To examine the rates of oocyte maturation and fertiliza...

Abnormal fertilization is responsible for reduced fecundity following thiram-induced ovulatory delay in the rat.

Brief exposure to some pesticides, applied during a sensitive window for the neural regulation of ovulation, will block the preovulatory surge of LH and, thus, delay ovulation. Previously, we have shown that a single i.p. injection of 50 mg/kg of thiram, a dithiocarbamate fungicide that decreases norepinephrine synthesis, on proestrus (1300 h) suppresses the LH surge and delays ovulation for 24...

P-76: Cytogenetic Investigation of Parthenogenetic Mouse Embryos Generated from In Vitro Activated Oocytes by Hydrostatic Pressure in The Presence of Calcium Ionophore and Ethanol

Background: The advances in cytogenetic techniques during the last few years have permitted not only the study of large populations of wild and domestic animals, but also the detection of chromosome anomalies in embryos. Chromosomal abnormalities are the most common cause of embryonic and fetal mortality in mammals. Most reports of chromosome anomalies in parthenogenetic embryos describe numeri...
